Histochemical GUS analysis
- Immerse
tissue in the GUS reaction buffer (2 mM
X-Gluc, 0.1 mM potassium ferricyanide,
0.1 mM
potassium ferrocyanide, 1 mM EDTA, and 50 mM NaPO4, pH 7.0).
- Vacuum
infiltrate briefly.
- Incubated
at 37°C for 4 to 16 h.
- After
incubation, clear seedlings methanol or 70% (v/v) ethanol to remove
chlorophyll for better visualization.
